Preparasi Rangka Utuh (Whole-Mount Skeleton) Vertebrata Kecil untuk Aktivitas Sains Sekolah

The first step in almost every investigation of skeletal phenotypes is analysis of whole-mount skeletal preparations. Whole-mount skeletal staining permits evaluation of the shapes and sizes of skeletal elements in their appropriate locations. The technique could be applied in thel teaching of skeletal system in high school. Teaching skeletal system would be more attractive and effective using real/preserved specimen compared to those using book/internet source.The aim of the research was to develop staining and clearing method of small vertebrate skeleton specimen using Alizarin Red S and produce teaching aid tools of skeleton system. The specimen were eviscerated, fixed, stained, cleared, and keep in glycerine solution. The spesimen is an alternative in high school science teaching and learning.
[Langkah pertama pada hampir semua penelitian fenotip rangka adalah preparasi rangka utuh. Pewarnaan kerangka keseluruhan memungkinkan evaluasi bentuk dan ukuran elemen kerangka di lokasi yang sesuai. Metode ini dapat diaplikasikan dalam kegiatan pembelajaran skeleton di sekolah menengah. Pengajaran sistem rangka akan lebih atraktif jika menggunakan spesimen asli atau awetan berbanding menggunakan buku/sumber internet. Pengamatan langsung memegang peranan penting dalam pemahaman siswa tentang fenomena biologi. Tujuan penelitian ini adalah untuk mengembangkan preparasi rangka utuh vertebrata kecil dengan pewarna Alizarin Red S sebagai suatu aktivitas sains di sekolah dan menghasilkan preparat skeleton sebagai sumber belajar sistem rangka. Spesimen diwarnai melalui beberapa tahapan yaitu dikuliti, difiksasi, diwarnai, dijernihkan dan disimpan dalam gliserin. Spesimen ini merupakan alternatif yang kreatif bagi pembelajaran sistem rangka di sekolah menengah.]
